Are there any set-up instructions for the 61-506 Tongue and Groove Edge Banding Set?

No written instructions are included but the setup for these bits is quite easy. Setting the profile to be centered in the stock is recommended but not necessary. I recommend running the workpiece that makes the tongue first, then using it to set the corresponding bit. In my experience, this works quite well.
